Diastaticus PCR test (one well isolated colony)
One of the most problematic Saccharomyces wild yeast strains is Saccharomyces cerevisiae var. diastaticus. This particular yeast strain can break down complex sugars such as dextrins (which normal S. cerevisiae strains can’t use) and cause overattenuated beers, resulting in bottle bombs. This PCR test will detect the presence of the STA1 gene that Diastaticus has, but normal S. cerevisiae does not. We highly recommend doing this test on well isolated colonies, as it is sometimes difficult to detect from a liquid culture.
